The 46-year-old salesman noticed a mole on his chest three and a half years ago that was found to be cancerous. Doctors removed the mole but didn't get all of the cancer. The cancer spread to other parts of his body, including his liver, where a tumor grew as large as a baseball. Parker took interferon and interleukin-2 to boost his immune system's ability to fight the cancer. The tumor shrank but didn't disappear. In au- gust, 1997, surgeons removed it, along with two-thirds of his liver. Last January, doctors discovered a new tumor on Parker's left adrenal gland(肾上腺). He received an ex- perimenta1 cancer vaccine at the University of Chicago Hospitals, but the vaccine did n' t stop the cancer from spreading to his right adrenal gland.

Immune therapy "ultimately will be a significant change in the way we treat a lot of different cancers," said Dr. Jon Richards of Lutheran General Hospital in Park Ridge, who is testing cancer vaccine on melanoma patients. "It will be an equal partner with the other three treatments in the next five to ten years." Several drugs that bolster the immune system have been approved, and vaccines are being tested in dozens of clinical trials, including several in the Chicago area. Many of the experimental vaccines have been tested on patients with advanced melanoma who have little chance of surviving with conventional treatments alone. Researchers also have begun doing work that could lead to vaccines to treat prostate, lung, colon and other cancers.

g friends with a dolphin. Well-wishers raised more than $10,000 to send Nikki Brice to Miami for treatment at a therapy center where handicapped youngsters are encouraged to swim with dolphins.

According to Professor Robert Kormos, one of the researchers, these results could revolutionize heart treatment. Although previous studies had indicated that there might be a benefit, this is the first study that has actually proved that stem cell therapy can help the failing heart work better.

All the patients in this study had hearts that could not pump blood properly. The scientists measured their ejection fraction (射血分数). It is a measure of heart performance; it measures how much blood is being pumped out by the left ventricle (心室).

Healthy people"s ejection fraction is about 55%. These patients had ejection fraction of under 35%. They all had by-pass surgery (搭桥手术) performed on them. Some of the patients had stem cells taken from their hip bones and injected into 25-30 sites in the damaged heart muscle. Six months later their ejection fraction rate was 46.1% while those who just had surgery but no stem cell injections averaged 37.2%. No side effects were reposed.

Heart failure is a common problem all over the world. In the U.K. alone about 650,000 people suffer from heart failure every year. As the number of people suffering from heart failure increases in the world in general, these findings are particularly significant.

Current treatments relieve the symptoms. This new stem cell therapy actually repairs the damaged muscle in the heart and has the potential of curing the disease.

A newly published AIDS study could open another front in the battle against HIV infection
by showing that gene therapy can be used to stop infected cells from spreading the deadly virus, researchers said.

In a test-tube experiment believed to be the first of its kind, researchers based at Children's Hospital of Philadelphia were able to block the operation of the" tat "gene that allows HIV to spread throughout the body from infected cells.

Up to now, genetic AIDS research has concentrated on finding ways to help healthy cells withstand the ravages of the HIV virus that infects an estimated 16, 000 new victims a day, mainly in the developing world.

But by working with human cells already infected with HIV, the team was able to reduce the tat gene's virus-replicating functions by 80 percent to 90 percent, according to findings published in the journal Gene Therapy.

That, researchers said, raises the possibility of a new gene therapy approach capable of supplementing the current drug-based treatment known as highly active anti-retroviral therapy, or HAART, which is used to stop HIV infection from becoming full-blown AIDS.

In recent studies, HAART has proved to be a costly drug regimen that poses serious side effects for HIV patients while delivering questionable results.

"This is proof of the concept that HIV replication could be inhibited by a genetic approach, though we're not at 100 percent yet, "said Dr. Stuart Starr, a study coauthor and chief of immunologic infectious diseases at Children's Hospital.

"Everyone thinks of an antiviral approach, or an immunologic approach to HIV. This adds another option into the equation that could become more important as other options prove not to be totally successful."

Key to the study was an artificially produced" antitat" gene provided by the Washington-based Research Institute for Genetic and Human Therapy.

Children's Hospital researchers used a mouse retrovirus to deliver the antitat gene into HIV-infected U-1 and ACH-2 cells, which were developed in the lab from the tissues of living HIV patients.

They found that when the antitat protein combined with the tat gene, it successfully inhibited the gene's operation without disturbing healthy cells or causing toxic side-effects.

The study, funded by a private foundation, also found that the introduction of the antitat gene prolonged the survival of immune-system cells called CD4 + T lymphocytes.

Start said researchers have entered preliminary discussions with a New England-based primate center ,where animal experiments could be carried out on infected macaque monkeys.

If animal experiments proved successful, the Children's Hospital team would hope to have a gene therapy treatment ready for human clinical trials in three to four years.
